Phenol:Chloroform:Isoamyl alcohol 25:24:1 is used in the extraction of DNA for the generation of DNA minisatellite fingerprints. It is also used for purifying RNA.

Description générale
• Cette solution contient un mélange de chloroforme pur, d’alcool isoamyle et de phénol saturé de Tris-EDTA, avec un pH stabilisé à 8,0
Applications
• La solution de mélange d’alcool isoamyle-phénol-chloroforme (25:24:1) est utilisée pour l’extraction des acides nucléiques, de l’ADN et de l’ARN
• Lorsque des acides nucléiques sont extraits avec la solution de mélange d’alcool isoamyle-phénol-chloroforme, une séparation des composants franche est obtenue.Les protéines présentes dans l’échantillon sont dénaturées et éliminées (collectées en phase organique ou en interphase), tandis que les acides nucléiques restent en phase aqueuse.Le chloroforme dénature les protéines et facilite la séparation des phases aqueuses et organiques, et l’alcool isoamyle réduit la formation de mousse pendant l’extraction.
• Elle est également utilisée pour générer des profilages d’ADN minisatellites
