Description

The Mouse Progranulin (GRN) ELISA quantitates Ms Progranulin in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms Progranulin. Principle of the method The Mouse Progranulin solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. La progranuline est la protéine précurseur de la granuline. Elle est également appelée proépithéline et constitue un facteur de croissance dérivé des cellules PC. Le clivage du peptide signal produit de la granuline mature. La granuline et le produit de clivage plus petit de la granuline régulent tous les deux la croissance cellulaire. La progranuline stimule la prolifération de nombreuses cellules épithéliales, favorise leur croissance indépendante de l’ancrage et confère la tumorigénicité épithéliale en cas de surexpression. Le gène de la progranuline /épithéline est multifonctionnel. En effet, il assure des fonctions essentielles dans l’homéostasie épithéliale, la fonction reproductive, immunologique et neuronale. La progranuline s’exprime de façon constitutive dans un certain nombre d’épithélia, en particulier dans la peau, le tube digestif et le système reproducteur. La progranuline s’exprime dans les cellules immunitaires in vivo et dans les neurones spécifiques du cerveau.
